Discussion Items:
  • Update on Mack progress
    • Have run into a hand homing issue.  Replacement of everything, and movement of hand to another instrument has the issue following the hand.  A new hand on Mack does not show this problem, but still unknown what is causing it.  Otherwise, everything calibrated on Mack (hand, intake pressures, carousal). 
    • Dynamic control of puck temperatures is good, but still only able to hold within 4 degrees of setpoint.  Most likely because temp sensor doesn't touch puck all that well, or this touching varies; thus final goal temperature is off.  Regardless, Brent thinks we are much better than the GG and Neo runs last year.
    • With the long weekend (Friday and Monday-Prez day) we will run non-stop mock missions through Mack (Flush, BAC, LARV,HAB, HAB-DA), before bagging on Tuesday and allowing science to run 'real' missions.  Deployment schedule has changed (see above link).

sh1 - 85C
DA - 70C
sh2 (laf, saf) - 30C

having said that, we might we drop the actual setting on the esp a couple degrees to remain on the safer side of the line (hotter is worse).  So Brent, use your judgement.  I know a setting of e.g. 28 for sh2 sould be hard to hit when working in a warmish room, but worth a try?

The outline of the simple  mission for the faux run is given below.  All sample vols should be 50 ml (sample for lysis and wcr).  All reagent lines, including sample) will be filled with flush fluid.  Exposures should be 160s 1x1, 40s 2x2, auto 2x2.  Should we #-out the auto exp since there should be no light, or just let it burn till time-out for kicks?

  •  Mike Parker contacted about piezoprinter problems (holding down membrane, cutting out disks) and is designing solutions.  Also will be approached about a puck holder.
  • New syringes.
    • Still not heard back from company
  • Screens and scintered frits
    • Will order 400 10um frits with frit packing tool. total cost $4300.00.  Order placed 2-14-08.  Here is the order and the quote.
    • We will order enough screens for 6 instruments.  (who is doing this?)
  • Puck Laser Etching
    • first half of pucks have returned from the etching, and SH1 tops and HV puck tops are mislabled.  Will investigate sending these back, as well as sending out the 2nd batch of lot2 pucks.  The first 'new' batch (lot3) has arrived from El Camino, Mike P is inspecting.  Has found they forgot to put hole in SAF bottoms.  Will return to get that done.  These pucks will go straight to etching before we  get them.

Discussion Items:

 Decided  to run thermal cycler all weekend with just water and bleach, to see if extra 'seasoning' of the tubing will lessen its inhibition of the PCR reaction.  Upon return next week, will run sample to test PCR effectivness.

ACTION:  Addie to look at heating profile times to make sure we are not cooking our reaction. (Addie)  Finished

Software

  • puck heating control
    • Brent has gotten calibration w/in 4degrees, but measuring puck temp is problematic.
  • Firmware 'lock-down'?
    • Thermal boards and Dwarf boards look fine, but the failing home sensor in the hand seems to generate a cascade of failures.  This will be fixed.(Brent)
    • GG and Neo will have old firmware and old Ruby.  Mack will use newer of each and these versions need to be kept separate.

Sphere and 4K DWSMS

  • Sphere update
    • No word or update from Wah Chang.
  • DWSM progress?
    • Doug believes he's found a pump that will push reliable, reproducible amounts of fluid.  Ordering motor to run it, and now must test that it pushes the correct volumes of fluids with no back-flow.
  • Battery boxes?  Will order 1 box (~$2200; acct 701312.04; Scott)

GUI Development

  • Mission script GUI introduction

Kevin has placed new GUI on the server here.  Rich Schramm available to do data parsing work; we want to utilize this by Sept deployment.

Chris P. and Brent need to address high volume issues for Sept deployment.
