Trial Deployment

• We will do a 'trial' deployment of new mooring to test before Cawthron.  Initially trying for week of Feb 27 using Shana Rae.  Scott leading this.  Jim Christman has been emailed to ask about dates (2-13).

Communications

• GSM does not support incoming data on their networks.  Brent has two possible solutions for data transfer that he's working on.

Deployment support

• Kevan and Chris P getting reagent list together.

 CANON Cruise

• Still unclear if drifter should be deeper than 25m.
• Will oxygen sensor be necessary?
• CANON meeting Feb 16 should answer these questions

3G planning

•  We will be doing dead-end filtration, although types of filters depends on volume and bugs.  All extractions have been on Durapore; haven't tested Supore.
• Bead beating is best for RNA and DNA extraction;  works best at room temp--have not tested yet at cold.
  • all testing has been on ribosomal targets.  Started testing functional genes, and 2 tested so far are in very low abundance.  Now waiting to get more from Vanessa.
  • All-Prep kits outperforms all others at overall nucleic acid extraction.  Does better at DNA than DNeasy, but RNeasy gets more RNA than all-prep.
• At Lab Automation next week look at two things:
  • filtration and lysing in same step.  CAn be done on same filter?
  • look for how people move fluids around.  Important for size fractionation, filtering of lysate, etc...
• Much discussion about who 'owns' the injection loop to analytical module.
  • What volume needs passing?  is it fixed, or could it be different depending on module needs?  must we 'condition' the sample before handing it off?  who cleans the loop?
• Our sample volume and how we break things really sets us apart.  After that, the extraction and handling is very similar to the rest of industry.
